Autoradiographic Studies on the DNA Metabolism in the Submaxillary Gland of Parathyroidectomized Chicken

Abstract
The author had studied autoradiograpbically on the effect of the parathyroidectomy on the DNA metabolism in the cells of the submaxillary gland of the chicken.
25 young experimental male chickens of weighing about 400 grams were divided into five groups, which composed 5 chickens. The DNA synthesis in the submaxillary gland cell were observed at interval of 6,10,18, and 32 hours after parathyroidectomy.
The animals were injected intraperitoneally a dose of 0.9 micro curie per gram body weight of ©øH-thymidipe at two hours before parathyroidectomy performed. The excised submaxillary glands were fixed in the 10 percent neutral buffer solution and sectioned of 15 microns with cryostatic apparatus. The autoradiography were prepared of NTB-3 Emulsion(Kodak Co., U.S.A.) and the slides were stained with Harris Hematoxylin.
Percentage from labelled nuclei per 1,000 nuclei in the submaxillary glands were calculated and the ratio was set as the radioactive indices.
The obtained results were as follows;
1. The DNA synthesis of submaxillary glands were decreased by parathyroidectomy.
2. The cell renewal rates of submaxillary gland were decreased in 10, and 18 hours after parathyroidectomy and recovered gradually.
3. These changes were showed in both mucous and serous cells in the terminal portion of submaxillary gland.
